Guptill was recalled to the New Zealand Test squad earlier this month for the tour of England, meaning Derbyshire needed a player to come in before Tillakaratne Dilshan links up with the county later in the summer.
Amla will make his Derbyshire debut in their home County Championship clash with Northants on May 10 and will play one more Division Two match as well as three games in the NatWest T20 Blast.
The 31-year-old, regarded as one of the best batsman in world cricket, has featured in 82 Tests scoring 6,757 runs at an average of 52.78 and a top score of 311 not out.
Amla is also an experienced limited overs batsman, scoring over 2,500 runs in 100 T20 appearances, with a high score of 88 not out, and he boasts a strike rate in international T20 cricket of 122.44, scoring at an average of 25.
