The British star slipped to third in the world rankings on Monday and, as expected, tournament organisers stuck by that list when announcing their seedings on Monday evening.
Murray is seeded three, behind top seed Novak Djokovic and Rafael Nadal, the man who climbed above him in the ATP rankings by winning the Masters 1000 title in Cincinnati on Sunday. He could therefore face Djokovic in what would be a rematch of last year's final, at the last-four stage.
The draw will be made at 1700 BST on Thursday.
Five-time winner Roger Federer is seeded only seventh after continuing his slide down the rankings.
The Swiss, who last won the US Open in 2008, is currently at his lowest ranking since October 2002.
He has subsequently been seeded behind last year's beaten semi-finalists David Ferrer (4) and Tomas Berdych (5), as well as the 2009 champion Juan Martin Del Potro (6).
Top-20 stars Jo-Wilfried Tsonga, Gilles Simon and Marin Cilic have all withdrawn from the tournament which means Dmitry Tursunov, ranked 35, squeezes into a seeding slot at number 32.
Meanwhile, the draws for US Open qualifying were made on Monday.
Briton James Ward will open against France's Jonathan Eysseric, while in-form Dan Evans takes on another Frenchman, Stephane Robert.
There are four Britons in the women's draw, led by Johanna Konta who has been seeded. She faces Australian Olivia Rogowska first up.
Samantha Murray also meets an Australian opponent in Arina Rodionova, Tara Moore takes on Catalina Castano, of Colombia, and former British number one Elena Baltacha faces a tough test against American Melanie Oudin, who made the quarter-finals of the main draw at Flushing Meadows back in 2009.
Players will need to win three matches to reach the main draw. Qualifying starts on Tuesday - when Ward, Konta and Baltacha will be in action - with the man draw getting under way next Monday.
