Batting first, Bangalore's hitman Chris Gayle propped up the innings with an unbeaten 92 from 58 balls, including 11 fours and five sixes, and led the Royal Challengers to 156 for five after they were asked to bat.
The West Indian was supported by captain Virat Kohli (24), who had to come on early after Tillakaratne Dilshan was bowled out for a duck by Mitchell Johnson in the third over.
Seam bowler Jasprit Bumrah claimed three for 32 to help reduce Bangalore to 80 for five in the 13th over but Gayle and Arun Karthik (19*) plundered an unbroken 76-run stand during the remainder of the innings.
Mumbai, victors in four previous fixtures at the ground, sent out the impressive opening pair of Sachin Tendulkar and Ricky Ponting - the top two Test run scorers of all time - to begin the chase and the duo registered a half-century partnership to calm the visiting fans.
However, the Little Master was run out by Jaydev Undakat for 23 in the eighth over and his Australian partner followed suit 14 balls later when he was stumped out by Arun Karthik off Murali's delivery after putting 28 runs on the scoreboard.
Dinesh Karthik (60) then came out and gave a semblance of hope for Mumbai as he took on the scoring responsibility and turned the tide in their favour with three consecutive sixes off Christian's bowling in the 18th over.
With 10 needed off the final over, Royal Challengers seamer Vinay Kumar stepped up. He conceded a single bye off the first delivery and then removed Karthik and Ambati Rayudu (18) with the next two.
Harbhajan Singh pushed a single to leave Kieron Pollard on strike with eight required and two balls remaining.
Pollard hit a full toss back over Kumar's head for four to take the contest down to the final delivery.
Kumar (three for 27) got his yorker on target and Pollard could only jam it for two runs on the leg side to ensure Bangalore emerged victorious.
Mumbai Indians can next get a chance to secure a first win in the 2013 IPL season when they take on Chennai Super Kings at the M. A. Chidambaram Stadium on Saturday night.
