The Birmingham boxer, who represented Great Britain at the 2008 Olympics, has impressed since turning professional in July and gets another chance on the 11-fight 'Christmas Cracker' bill.
Yafai took just 52 seconds to dispose of Pio Nettuno when he fought on the Carl Froch - Yusaf Mack undercard, taking his record to 5-0 with four stoppages.
"Everyone is talking about Kal Yafai right now and Perez represents a significant step up in class for him," said promoter Eddie Hearn.
"We are fast tracking him to the top. This is his sixth fight in as many months and we expect him to challenge for title honours early next year."
Perez won the Spanish title with a third-round knockout of European title challenger Ivan Pozo in October.
The 28-year-old has won 11 of his 16 fights but was knocked out when he fought Ali Hallab for the WBO European super bantamweight title in May.
