Klose, 34, scored a goal in the first half but confessed to having committed a handball in a display of professionalism that was widely applauded by both sets of fans.
As the chief striker for The Eagles, the German has scored 16 goals from 31 appearances in two seasons but fitness concerns and a lack of support has made it difficult for him to replicate the goalscoring form he enjoyed back in the Bundesliga in previous seasons.
Petkovic was critical of his side's performance against Napoli and demanded the players not rely solely on the German international.
"Klose can't always be the one to resolve situations. The others have to take responsibility too," Petkovic told Rai Sport after the game.
"We must remain calm and try to introduce other strikers in future.
"Today [Wednesday] certain incidents went against us, like Klose who scored and confessed he had handled it.
"We had been dangerous early on and Napoli were afraid, but after their opening goal everything changed.
"Hernanes had a problem with his leg, so was not 100 per cent tonight, but Napoli deserve compliments for pressuring him well."
"Miroslav did well to admit he touched the ball with his hand, but we had to take advantage of this occasion as something positive, and instead we slowly switched off."
Klose's sportsmanship went down well with Napoli captain Paolo Cannavaro, who said of the experienced striker: "He deserves a prize."
Napoli coach Walter Mazzarri, on the other hand, was full of praise for his side's performance in their fifth win in the season. They are now second in the Serie A table and look the only side capable of challenging Juventus for the title at this early stage.
"The whole team played a great game and I am happy with everyone at Napoli tonight," he said.
"I told the side that we could lose this evening, because Lazio are a great side, but the important thing was to see the right attitude and character on the field. That is what I saw tonight.
"Valon Behrami did particularly well, as he won back possession dozens of times and ran so hard to help the team."
Napoli next play Sampdoria while Lazio host Siena on Sunday.
