The Spanish Under-21 star has been heavily linked with a move to Premier League champions Manchester United in recent weeks, after struggling to displace established Barcelona stars like Andres Iniesta and Xavi from the starting line-up last season.
However, Poyet told ESPN that the Catalan side would be passing up on a player with great potential if they allowed Thiago to leave the Camp Nou.
"When I heard about a possible move away, I couldn't believe it - I never thought it would actually happen," he said.
"I see Thiago as the replacement for Xavi, Iniesta and Cesc Fabregas. He is someone who could make history at the club."
Echoing Poyet's comments, Barcelona defender Gerard Pique hopes the club will do whatever it takes to keep the 22-year old.
"[Thiago] is a top level player and we cannot let him escape," he stated to RAC1.
"We must do everything possible to keep him."
