The Blaugrana have enjoyed a good start to the season and are currently leading the La Liga standings with 22 points from eight games, 12 points ahead of their next league opponents Rayo, who are in 12th place.
However, despite the points disparity between the two sides, Vilanova claims that the Matagigantes will prove to be difficult to beat and credited Rayo coach Paco Jemez for improving the team.
"The trip to Rayo is the toughest trip so far," Vilanova was quoted as saying by the official Barcelona website.
"Paco Jémez has done a great job. It won't be easy for us to come out with the ball, because they are a very adventurous side.
"Vallecas is never an easy ground to visit and on Saturday they won't just sit back and wait for us - they'll go toe to toe with us".
Vilanova was also quick to remind his players that they had to remain consistent in order to wrest the La Liga title from champions and arch-rivals Real Madrid come the end of the season.
"I am very happy with the players' performances and I have no complaints," Vilanova added.
"Winning titles is important, but it's maybe even more so to have the confidence that the players will be in there till the end of the season."
Barcelona will have pleasant memories of their visit to the Estadio Teresa Rivero last season where they romped to a comfortable 7-0 win over Rayo.
