Agger and Skrtel have struck up a fine partnership in the heart of the Reds backline, even consigning club vice-captain Jamie Carragher to the bench for much of the season.
And Enrique, who moved to Anfield last summer and has made the left-back spot his own, believes both of them are the best in the league at the moment.
"I play with amazing players," Enrique said on the club's official website. "For me, I wouldn't change our centre-backs for any centre-backs in the league.
"They are both playing amazingly."
Enrique went as far as to declare that the Reds have arguably one of the best defences in the Premier League, after hailing both right-back Glen Johnson and goalkeeper Pepe Reina.
"We have one of the best right-backs in the world, because Johnno [Johnson] is really good, and one of the best keepers [Reina], and we have amazing players in front who help us defence."
The Spaniard was keen to stress the importance of keeping things tight at the back and racking up the clean sheets, although he admitted picking up three points was more important at the end of the day.
"Clean sheets are really important because if you keep one, you have one point for sure," Enrique added. "But if we won each game 6-5, I wouldn't care.
"The most important thing is winning the games, but I'm really happy when we keep clean sheets.
"We are doing well. We defend and attack as one - we play together. I'm happy because as a team we're doing well - only Manchester City are ahead of us in clean sheets.
"We have to keep doing this and score more goals."
Liverpool are next in action on Sunday when they take on Brighton in the FA Cup.
