Both clubs have already qualified for the last 32, with Hannover dropping one point in five games while Levante's solitary defeat came against the German side earlier this season.
With FC Twente and Helsingborg out of the qualifying picture early on, the group will be decided by Thursday's encounter in Valencia.
Hannover are currently a point clear of their opponents, and they know that a draw would see them progress as group winners. Levante, meanwhile, need victory to finish top.
Levante have made an impressive debut in European competition this season, winning both their home games to underline the challenge facing Hannover.
Since the last round of European fixtures, Levante suffered a 4-0 defeat against La Liga title favourites Barcelona, but a 1-1 draw against Celta Vigo last time out will have gone some way to restoring confidence levels.
But it has not been all sweetness and light for Hannover, whose recent Bundesliga performances included a 5-0 drubbing by runaway leaders Bayern Munich.
An indifferent league position of 12th confirms the Germans have produced their best form in Europe this season, which will be a fact not lost on Levante.
Hannover will head to Spain on the back of a 2-1 league loss to 10-man Mainz.
"We made technical mistakes and we didn't work ourselves enough chances in front of goal, despite having more players," Hannover captain Steven Cherundolo told the club's official website www.hannover96.de.
"You can't win away from home when you play like that."
Coach Mirko Slomka has almost a full-strength squad at his disposal, with only long-term absentees Leon Andreasen (knee ligaments) and Felipe (hip) not in contention.
Levante's Hector Rodas has recovered from a thigh injury and is likely to cover for the suspended Sergio Ballesteros in the centre of defence, while full-back Juanfran is also back in full training after a back problem.
Midfielders Dariusz Dudka and Pedro Rios continue to work their way back to fitness along with striker Obafemi Martins, with the latter ineligible for the competition in any case.
