Tampines, who qualified for the competition for the second year running by virtue of being Great Eastern-YEO's S.League champions, will have a chance to make amends for failing to make it to the knockout round next year.
However, they will have to negotiate through some tricky ties after being handed a difficult group which includes Selangor, who finished third in the Malaysia Super League this year but qualified for the AFC Cup as runners-up LIONSXII are ineligible to represent Malaysia.
The clashes against the Red Giants will also be the second time in two years Tampines face Malaysian opposition, after taking on Terengganu this year.
Also looking to progress from the group are India's East Bengal and V-League outfit Saigon Xuan Thanh.
The Stags' S.League rivals SAFFC, who qualified after winning the RHB Singapore Cup, were put in Group E and will face Hong Kong's Kitchee, who played against Tampines earlier this year, Indonesian giants Semen Padang and Churchill Brothers of India.
Meanwhile, Malaysia Super League champions Kelantan, who also won the Malaysia Cup and FA Cup this term, will be looking to repeat their run to the quarter-finals by progressing from Group G, which also includes Ayeyawady United, Da Nang and Maziya.
Group F is the final group involving Southeast Asian sides and will see Myanmar's Yangon United lock horns with Indonesian outfit Persibo Bojonegoro, as well as Sun Hei of Hong Kong and Maldives' New Radiant.
